Despite COVID-19 lockdowns, gradual restarting of the global economy has rapidly increased air quality index (AQI) values. With over 99 % world population living in areas exceeding guidelines, pollution is more so a threat to our health. Within particulate matter, major pollutant, lies polycyclic aromatic hydrocarbons (PAH). While recent studies explore link between and pigmentation disorders, molecular mechanisms responsible for this alteration remain largely unknown. To challenge hypothesis that exposure PAH leads an increase abnormal pigmentation, we have utilized vitro vivo assays. In vitro, novel 2D 3D co-culture assays were developed analyze pigment production, transfer total melanin content human keratinocytes melanocytes. Following that, bulk RNA-sequencing was also carried out on isolated melanocytes post possibly elucidate mechanism behind phenomenon. vivo, mouse model bearing epidermal generated investigate effect exposure.
